I could never pick a "best fighting game", but I have a lot to say about this topic
Tekken Tag is much better than Tekken 3, and every other tekken in the world! Balance, characters, and overall fun is much better in Tag. 3 was a little silly because of all the broken stuff. (King gets 65 damage off a 2,1. Ogre gets free waning moon after blocked hunting hawk, Jin gets a free EWGF after d/f+2, etc.) I haven't been a real tekken fan since Tag, but I can play DR and 6 decently.
This generation has Super Street Fighter IV, which is easily one of the best street fighter games ever, along with SF3: 3rd Strike and Super SF2 Turbo.
MVC2 is a GREAT game, as well. It's hated for being "stupid" and "cheesy" by people who don't get it, but it's the fastest, most technical fighting game out there.
I really think it's funny that fighting games are misunderstood as shallow and simplistic, when in reality they are incredibly deep and difficult to play properly. I guess it makes sense, though, because a spectator can see the punches and kicks and completely miss the mind games, strategy, zoning, and hand technique required to play!

Shouldn't this thread be in a more general section? There are fighting games on every console, and not just the Xbox 360
